Any flat route to Adelaide for you would entail a much longer route. The bit of extra time the slow running on the direct route would be far less than the extra time going the flat route.
What is wrong with your rig that you want to dodge hills? Is the tug under-powered or are you pulling too large a van? I suggest you get used to hills if you are coming over to the east coast.

There are a couple of slow drags, with the first one after Callington, and one as you get near to Stirling being the worst in my opinion. Take them easy, and you would be fine
That said, it may be a bit flatter going up to Mannum and coming from there, or down to Victor Harbor and coming up from there, but then you face roads that are windy and narrow and have there own set of issues. I wouldn't bring a van via Mannum and Chain of Ponds, especially anything over a single axle due to the windiness.
At least on the SE Freeway from Murray Bridge, you have 2 lanes all the way, so you will not need to belt along, as traffic can get past easily. Also much shorter (only about 75 kms), easier for RAA support if you have issues, and a better road.
Unfortunately. like the east coast, if you want to come to Adelaide, and are coming from the east, you have to cross the Mount Lofty Ranges.

Shouldn't have too much trouble up the direct route only towing 900kgs.
Towed over that piece of road several times with 3000kg on the back. Admittedly using a Ranger but is a fairly easy route.
