As the 5th and 6th gears in the 6 speed auto are both overdrive I'd personally be towing in 5th. While the 6 speed box doesn't have a 1:1 ration available 5 is reasonably close at 0.87:1 ration. While the revs will be higher than in 6th the engine will be turning over easier and there will be less strain on the torque converter as it will reduce gear changes and torque converter continually unlocking and locking again. We tow with a TS RWD diesel and pull a similar size van. Tows like a dream, but we use auto.

We had the extra seats removed, and so have a wet storage area in the back, where the seats were. We had a luggage barrier installed, and the dogs travel safely behind it in the back.
However, I must say, while we get great economy, its nowhere near what they advertise - got close once on the Hay Plain in NSW - dead flat, dead straight, not towing and no air con.
Towing we get about 11-12 litres / 100 kms- depends though.
